1. Biofilm Adhesion Capacity (Key Performance Indicator)
- Biofilm attachment capacity = Protected surface area (determined by media design) × Biofilm attachment per unit area (media property-dependent)
2. Surface Characteristics
a) Surface Texture
- Rough surfaces demonstrate faster biofilm formation rates
b) Surface Charge
- Optimal microbial growth occurs when media surface exhibits positive charge (microorganisms typically carry negative charge)
c) Hydrophilicity
- Hydrophilic media surfaces promote microbial attachment as microorganisms are hydrophilic particles

3. Hydraulic Properties
a) Porosity
- Higher porosity media is preferred (greater protected volume)
b) Geometry
- Shape and dimensions significantly influence hydraulic/airflow patterns
c) Fluidization Characteristics
- Optimal specific gravity range: 0.97-1.03 g/cm³
- Achieves effective fluidization with minimal aeration/mixing energy
4. Biofilm Maturation Assessment
Visual Inspection Criteria:
- Uniform biofilm distribution on media surface
- Increasing biofilm density toward media core
- Darkening media coloration
Microscopic Examination Criteria:
- Dense biofilm structure
- Diverse microbial population
- Dominance of sessile ciliates (Vorticella, Epistylis)
- Presence of rotifers and free-swimming ciliates
- Appearance of higher trophic organisms indicates mature biofilm
